Dies trägt maßgeblich zum Verständnis bei, wie eine Immunantwort aufgebaut, erhalten und - wenn nötig - wieder abgeschaltet wird, um den Organismus vor Krankheitserregern zu verteidigen, aber auch dem Aufbau von schädlichen Autoimmunerkrankungen - Überreaktionen des Immunsystems - vorzubeugen.
Unsere Forschungsgruppe "Intravitalmikroskopie von Infektion und Immunität" (INMI) hat sich auf die Multiphotonenmikroskopie spezialisiert, um das Zusammenspiel von Wirt und Pathogen im lebenden Gewebe zu verfolgen. Unser Fokus gilt hierbei der Frage, wie ein Pathogen auf den Stress reagiert, dem es aufgrund einer Immunantwort ausgesetzt ist. Indem wir Infektionserreger mit fluoreszierenden Reportersystemen (vom Erreger produzierte Farbstoffe, die auf biologische Veränderungen reagieren) ausstatten, können wir Prozesse wir Zellteilung und Proteinproduktion, aber auch die Aktivität von stressinduzierten Enzymen im lebenden Pathogen bestimmen.
Die Vermessung dieser Parameter während der Infektion erlaubt uns Rückschlüsse darauf, wann und unter welchen Bedingungen die Immunantwort am effektivsten ist. Da die biologischen Prozesse im Pathogen Einfluss auf seine Empfindlichkeit gegenüber Antibiotika haben, können wir auch herausfinden, wie die Immunantwort am besten mit therapeutischen Maßnahmen unterstützt werden kann.
