Einfluss des peripheren Nervensystems auf die Dynamik und Plastizität von Tumor-Immunzell Interaktionen
Laufzeit: 01.04.2022 bis 31.03.2027

Die molekulare Identifizierung von immuninhibitorischen Rezeptoren auf T-Zellen und die Entdeckung, dass die Antikörper-vermittelte Blockade dieser Rezeptoren die antitumorale Immunität verstärken kann, stellt einen bedeutenden, praxisverändernden konzeptionellen Fortschritt dar, der die Krebsmedizin im letzten Jahrzehnt revolutioniert hat. In diesem Projekt bauen wir auf neuen Erkenntnissen auf, dass das Nervensystem an der Regulierung sowohl der Funktionen von Immunzellen als auch von Krebszellen beteiligt ist. Periphere Neuronen führen eine bidirektionale interzelluläre Kommunikation durch eine Fülle von chemischen Botenstoffen durch, die den Wirt auf extrinsische und intrinsische Gefahren aufmerksam machen und helfen, Schutz- und Regenerationsreaktionen zu koordinieren. Wir werden mit Gruppen zusammenarbeiten, die an der Schnittstelle der neurobiologischen Hirnforschung und der zellulären Immunologie arbeiten, um die dynamischen und wechselseitigen Interaktionen zwischen Neuronen, Immunzellen und Krebszellen im Verlauf der malignen Progression und als Reaktion auf therapeutische Interventionen zu untersuchen. In unserer hochgradig interdisziplinären Arbeit werden wir die übergreifende Hypothese untersuchen, dass periphere Neuronen immunsuppressive Aktivität in der Tumormikroumgebung durch chemische Botenstoffe ausüben, die als zielgerichtete Neuro-Immun-Checkpoints für verbesserte kombinatorische Strategien zur Krebsimmuntherapie fungieren. Die gentechnisch hergestellten experimentellen Mausmodelle für primäre und metastasierte Melanome, die wir über die Jahre entwickelt und sorgfältig charakterisiert haben, stellen eine solide Basis dar, um diese Hypothese zu testen. Wir werden neuartige und innovative experimentelle Werkzeuge, die von Hirnneurobiologen entwickelt wurden (wie z.B. genetisch kodierte Sensoren und Designer-Rezeptoren, die eine optogenetische und chemogenetische Modulation neuronaler Funktionen ermöglichen), auf die Untersuchung peripherer neuronaler Funktionen in der Tumormikroumgebung anwenden, ein Unterfangen, das in anderen Programmen nicht finanziert werden würde. Dies wird uns erlauben, die Dynamik und Plastizität von Tumor-Immunzell-Interaktionen mit Hilfe von Intravitalmikroskopie, konfokaler Mikroskopie und Durchflusszytometrie zu untersuchen, die wir in unserer Gruppe zusammen mit unseren Partnern in der Immunologie routinemäßig durchführen. In unserer Arbeit wollen wir letztlich Neuropeptide und Rezeptoren mittels mit scRNAseq-Methoden identifizieren, die von peripheren Neuronen freigesetzt werden und die Anti-Tumor-Immunität einschränken sowie tumorfördernde Entzündungsreaktionen unterstützen. Wir werden experimentelle in vivo-Arbeiten in Mausmodellen mit histopathologischen und in vitro-Untersuchungen von menschlichen Melanomen kombinieren, um unsere Ergebnisse zu validieren und zu übersetzen. Die Entdeckung von "Neuro-Immun-Checkpoints" wird einen weiteren bedeutenden konzeptionellen Fortschritt darstellen, der das Potenzial hat, Kombinationstherapien bei Krebserkrankungen weiter zu verbessern. Unsere strategische Position zwischen exzellenter Forschung und Patientenversorgung an der Klinik für Dermatologie (zu der auch das Labor für Dermatopathologie gehört) bietet einen idealen Rahmen, um das translationale Potenzial unserer Arbeit in Zukunft zu erforschen.

Abgeschlossene Projekte

Mechanismen und therapeutische Beeinflussung kognitiver Funktionsstörungen bei Patienten mit fortgeschrittenem Hautkrebs unter Systemtherapie
Laufzeit: 01.12.2022 bis 30.11.2023

Neue Möglichkeiten in der Diagnostik und Therapie haben die Versorgung von Patienten mit (Haut-) Krebs in den vergangenen Jahren signifikant verbessert. Heute kann das Überleben auch bei invasiv wachsenden und metastasierenden Tumoren deutlich verlängert werden. Deshalb gewinnt ein besseres Verständnis nicht nur der körperlichen, sondern auch der psychischen Langzeitfolgen der neuen Behandlungsformen von Krebserkrankungen zunehmend an Bedeutung. Im Rahmen des hier beantragten Clinician-Scientist-Projektes soll dabei speziell auf den Erhalt der kognitiven Gesundheit bei Patienten mit (Haut-) Krebs eingegangen werden. Die Erforschung kognitiver Funktionen wie Lernen und Gedächtnis stellt einen international sichtbaren und interdisziplinären wissenschaftlichen Schwerpunkt an der Universität in Magdeburg, dem DZNE-Standort Magdeburg und dem Leibniz-Institut für Neurobiologie dar. Über die Auswirkungen einer monate- bis jahrelangen Therapie mit Signaltransduktions-Inhibitoren oder Immuncheckpoint-Blockern auf höhere Hirnfunktionen wie Lernen und Gedächtnis bei Patienten mit fortgeschrittenem Hautkrebs ist bislang wenig bekannt. Dies soll im Projekt mit neuen und innovativen Methoden in Kooperation mit AGs der kognitiven Neurowissenschaften untersucht werden. Dabei sollen auch Ansätze erprobt werden, um die Beeinträchtigung kognitiver Funktionen zu minimieren. Weiterhin sollen digitale Applikationen entwickelt werden, die es erlauben, bei der klinischen Betreuung von Patienten neben etablierten biomedizinischen Parametern auch kognitive Fähigkeiten und weitere psychosoziale Faktoren für Therapie-Entscheidungen zu berücksichtigen. Schließlich sollen präklinische experimentelle Modellsysteme etabliert werden, mit denen die Ursachen kognitiver Funktionsstörungen unter Signaltransduktions-Inhibitoren oder Immuncheckpoint-Blockern in präklinischen Modellen näher untersucht werden können.

Gq/11-gekoppelte GPCR Signalwege in der Pathogenese und Therapie des malignen Melanoms
Laufzeit: 01.02.2019 bis 31.10.2022

Die fehlerhafte Expression oder Aktivierung von G-Protein gekoppelten Rezeptoren (GPCR) und G-Proteinen spielt bei der Entstehung vieler Krebserkrankungen eine zentrale Rolle. Durch die Entdeckung somatischer Gnaq-Mutationen in blauen Nävi der Haut und bei Aderhautmelanomen wurde die Bedeutung des GPCR-Gaq-Signalwegs für die Entstehung melanozytärer Neoplasien deutlich. Ziel unseres Antrages ist es, die Rolle des Gaq/11-Signalweges in der Pathogenese des malignen Melanoms besser zu verstehen. Unsere Hypothese ist, dass die fehlerhafte Aktivierung dieses Signalweges abhängig vom Gewebe die Entstehung von Tumoren fördern kann. Mit Hilfe unserer Arbeit wollen wir neue Einblicke in die molekularen Mechanismen, warum und wie eine mutierte Gaq/11-Signaltransduktion das Wachstum und die systemische Ausbreitung bestimmter Melanom-Subtypen antreibt, gewinnen. Dabei hoffen wir eine Grundlage für die Entwicklung neuartiger therapeutische Ansätze u.a. für die Behandlung des Aderhautmelanoms zu finden.

Regulation von CD4+ T Zell Effektorfunktionen in Melanomen
Laufzeit: 01.01.2018 bis 30.06.2022

Jüngste Erkenntnisse zeigen, dass CD4+ T-Zellen wesentlich zu einer effektiven Tumorimmunabwehr beitragen können, aber auch an der Gewebehomöstase und- regeneration sowie an der Tumorpromotion beteiligt sind. Ziel unseres Forschungsvorhabens ist es, die molekularen und zellulären Mechanismen besser zu verstehen, wie die Phänotypen und Effektorfunktionen von  CD4+ T-Zellen im Tumorgewebe in vivo reguliert werden. Hierzu verwenden wir adoptive T Zell Protokolle in unseren experimentellen Mausmelanommodellen. Ein Fokus unserer Arbeiten ist die immun-regulatorische Rolle von neutrophilen Granulozyten lokal im Tumorgewebe und systemisch.

Die Bedeutung des inflammatorischen Mikromilieus für die Krebsentstehung (Stipendiat: Dr. Andreas Braun)
Laufzeit: 01.01.2019 bis 30.06.2021

Das Melanom stellt trotz neuer Therapieverfahren weiterhin die Hautkrebsform mit der höchsten Letalität dar. Haupttodesursache ist die Fernmetastasierung des Melanoms. In eigenen Vorarbeiten wurde das Hgf-Braf-Cdk4 Mausmodell etabliert, welches eine transgene Hgf-Überexpression, eine Cdk4-Keimbahnmutation sowie ein in melanozyten spezifisch aktivierbare BrafV600E-Mutation kombiniert. Dieses zeigt nach Aktivierung der Braf-Mutation eine ausgeprägte Fernmetastasierung in die Lunge. Auffällig war eine perivaskuläre Ausbreitung der Melanomzellen entlang der äußeren Gefäßwände. In dieser Arbeit soll der Einfluss einer UV-induzierten Entzündung in der Haut auf die Metastasierung im Hgf-Braf-Cdk4 Mausmodell untersucht werden. Besonderes Augenmerk soll dabei auf die Entzündungs-getriebene Interaktion von Melanomzellen mit Endothelzellen gelegt werden. Letztendliches Ziel ist es, molekulare Mechanismen der Metastasierung von Braf-getriebenen Melanomen zu entschlüsseln und die Bedeutung der so gewonnenen Erkenntnisse für das humane System unter anderem in histopathologischen Untersuchungen auszuloten.

Onkolytische Immun-Virotherapie des Melanoms
Laufzeit: 01.10.2017 bis 31.05.2021

Melanome sind bösartige Tumore, die als Folge einer UV-induzierten genomischen DNA-Schädigung in pigmentproduzierenden Melanozyten der Haut entstehen. UV-induzierte Entzündungsreaktionen unterstützen die Proliferation entarteter Zellen und fördern ihre Wanderungseigenschaften, ihr invasives Wachstum und ihre Fähigkeit zur Ausbildung von Metastasen in anderen Geweben (Bald…Tüting, Nature 2014). Die meisten Melanome der Haut werden frühzeitig erkannt und durch eine adäquate chirurgische Resektion geheilt . Bei einem Teil der Patienten entstehen jedoch Metastasen in inneren Organen, die bis vor kurzem fast immer zum Tod führten. Seit kurzem stehen neue Therapieverfahren wie die gezielte Inhibition von Wachstumsfaktor-Signalkaskaden oder die Blockade von immunregulatorischen Rezeptor-Interaktionen zur Verfügung, mit denen bei vielen Patienten eine Stabilisierung des Krankheitsverlaufs und bei einigen Patienten auch lang anhaltendeTumorregression erreicht werden können. Die Überwindung der primären und sekundären Therapieresistenz stellt aktuell eine der wichtigsten Herausforderung für die Forschung auf diesem Gebiet dar. Als eine attraktive Möglichkeit für zukünftige innovative Kombinationstherapien gilt gerade von der FDA für Patienten mit Melanom-Metastasen zugelassene onkolytische Virus T-VEC. Onkolytische Viren führen zu einer bevorzugten Infektion und Zerstörung von Tumorzellen. Dabei stimulieren sie gleichzeitig eine gegen Tumorzellen gerichtete Immunabwehr. Die onkolytische Infektion von Tumorzellen wird jedoch durch das anti-virale Typ I Interferonsystem begrenzt , das in verschiedenen Melanomzellen stark variabel aktivierbar ist. Unsere eigenen Vorarbeiten deuten darauf hin, dass die Aktivierbarkeit desTyp I Interferonsystems in Melanomzellen und die damit verbundene Sensivität gegenüber einer Infektion mit onkolytischen Viren invers mit dem melanozytären Differenzierungsgrad korrelieren. Wesentliches Ziel dieses Forschungsvorhabens ist die Prüfung der Hypothese, dass eine onkolytische Infektion und Lyse gerade bei differenzierten Melanomzellen wirksam ist und zur Aktivierung einer zytotoxischen T- und NK-Zell-Antwort gegen diese oft Immunzell-armen Tumoren beitragen kann. Die so induzierte anti-tumorale Immunabwehr kann durch eine zusätzliche Blockade von immunregulatorischen PD1/PD-L1 Rezeptor-Interaktionen weiter verstärkt werden. Im ersten Teil des Forschungsprojektes werden wir in einem erweiterten Kollektiv ausgewählter humaner Melanomzellen den Zusammenhang zwischen dem Differenzierungsstatus, der Aktivierbarkeit des Typ I interferonsystems und der Sensivität gegenüber einer Infektion mit dem Typ I-sensitiven onkolytischen Virus SFV VA7 in vitro vertiefend untersuchen. Weiterführend werden wir die molekularen Mechanismen ergründen, die diesen Zusammenhang erklären können. Im zweiten Teil des Forschungsprojekts werden wir diese Aspekte in vivo in dem von unserem Labor etablierten genetischen HGF-CDK4(R24C)Maus-Melanommodell addressieren und die Fähigkeit von onkolytischen Viren für die präferentielle Infektion von differenzierten Melanomzellen mit einer schwachen Aktivierbarkeit des Typ I Interferonsystems untersuchen. Auf diese Weise soll die primäre Resistenz dieser Melanome gegen eine Blockade immunregulatorischer Rezeptoren überwunden werden. Diese experimentellen Untersuchungen tragen letztlich zu einem besseren mechanistischen Verständnis der Wirkungsweise von onkolytischen Viren als Teil von neuen, innovativen Kombinationstherapien für Patienten mit fortgeschrittenem Melanom bei. Daraus ergeben sich wichtige therapeutische Implikationen für die Stratifizierung und Entwicklung von personalisierten Ansätzen der Krebsbehandlung.

Die Rolle des Arylhydrocarbon-Rezeptor Signalweg in metastatischen Progression und Therapieresistenz des Melanoms (Stipendiatin: Dr. Miriam Mengoni)
Laufzeit: 01.02.2020 bis 01.01.2021

Melanomzellen zeichnen sich durch eine ausgeprägte Plastizität aus, die es ihnen erlaubt, sich an wechselnde Umstände schnell anzupassen. Hierdurch können Melanomzellen Fernmetastasen bilden und Resistenzen gegenüber Systemtherapien entwickeln. Entzündungsprozesse spielen für diese Plastizität eine grundlegende Rolle. Der Arylhydrocarbon-Rezeptor (AhR) ist ein ligandenbindender Transkriptionsfaktor, der Entzündungs- und Differenzierungsvorgänge in der Haut beeinflusst. UV-Strahlung, der wichtigste Risikofaktor in der Entstehung des Melanoms, erzeugt aus der Aminosäure Tryptophan den AhR-Liganden Formylindolo(3,2-b)carbazol (FICZ) und aktiviert dadurch indirekt den AhR. In eigenen Vorarbeiten hat die Antragstellerin einen funktionellen Effekt des AhR in dem im Labor etablierten Hgf-Cdk4 Mausmodell nachweisen können. In diesem Projekt wird die Hypothese verfolgt, dass die Aktivierung des AhR-Signalweges die Pathogenese des Melanoms beeinflusst. Hierzu sollen Experimente in der Zellkultur und im Mausmodell in vivo durchgeführt werden.

Translational studies of the targeted and immune-therapy combinations in genetic engineered BRAF wild type melanoma models
Laufzeit: 01.08.2016 bis 31.07.2020

Background and objectives

In recent years significant breakthroughs in the treatment of metastatic melanoma have been achieved. Small molecule signal transduction inhibitors (e.g. directed against mutated BRAF or MEK kinases) and immunotherapies (e.g. adoptive T cell therapies or antibody-mediated blockade of immune checkpoints) have been shown to prolong the survival of patients with advanced disease. However, primary or secondary resistance limits the long-term efficacy of these therapeutic approaches. In this project we investigate in appropriate preclinical mouse models for melanoma multimodal regimens that effectively combine signal transduction inhibitors with immunomodulatory agents for the treatment of melanoma and try to gain fundamental insights that help to rationally design such combination treatment protocols exploiting the Genentech/Roche drug portfolio.
Aims
1. Evaluate the impact of Genentech/Roche MAPK inhibitors (e.g. MEKi, ERKi alone and in combination) on the growth and phenotype of HGF-CDK4(R24C) mouse melanoma cells as well as their interaction with immune cells in vitro and in vivo.
 
2. Optimize protocols that are able to enhance the efficacy of our established immunotherapies (e.g. innate immune stimulation and adoptive T cell therapy) in the HGF-CDK4(R24C) mouse melanoma model with Genentech/Roche immunomodulatory agents (e.g. IFNa, aPDL1 mAb, IDOi alone and in combination).
 
3. Identify strategies that effectively combine Genentech/Roche MAPK inhibitors and immunomodulatory agents in the HGF-CDK4(R24C) mouse melanoma model.
 
Perspective
The long-term goal of our work is to provide valuable information that can facilitate the clinical translation of effective combination therapies in ongoing and future trials in order to improve melanoma patient care. As MAPK pathway inhibition and novel immunotherapies also play a critical role in other tumor entities, the expected results of our experiments will likely also have implications beyond melanoma. In addition, we may delineate strategies to prevent unexpected interferences between the different treatment modalities regarding the mechanisms of therapy resistance.

Experimentelle Entwicklung von Strategien für die effektive Kombination von T Zell Immuntherapien mit der Hemmung von Tumorwachstums-fördernden Signaltransduktionswegen zur Therapie des Melanoms
Laufzeit: 01.11.2015 bis 31.10.2018

Die Entwicklung von Strategien für die effektive Kombination von T Zell Immuntherapien mit der Hemmung von Tumorwachstums-fördernden Signaltransduktionswegen zur Therapie des Melanoms stellt eine der aktuell wichtigsten klinischen Herausforderungen dar. Das wesentliche Ziel dieses Forschungsvorhabens ist die experimentelle Evaluation solcher Strategien in state-of-the-art präklinischen, genetischen Mausmodellen. In den geplanten Arbeiten werden wir die generelle Hypothese prüfen, dass die von Interferonen und CD8+ T Zellen getriebene zytotoxische Immunabwehr im Mikromilieu von Melanomen durch physiologische, protektive Mechanismen im Gewebe und durch die immunsuppressive Aktivität von Melanomzellen limitiert wird. Zu diesen gegen-regulatorischen Mechanismen zählen unter anderem die Rekrutierung von myeloiden Immunzellen in geschädigtes Tumorgewebe, die Stimulation von PD1/PDL1 immun-inhibitorischen Rezeptor-Interaktionen, und die Generation eines immunsuppressiven Milieus durch die Aktivität onkogener Signalwege in Tumorzellen. Die experimentellen Arbeiten zielen auf eine Interferenz mit diesen Mechanismen ab und bestehen aus drei Teilen mit den folgenden Zielen:

(i) Charakterisierung der Rolle von Typ I Interferonen für die funktionelle Regulation von anti-tumoralen CD8+ T Zell Antworten; (ii) Etablierung von in vivo Biolumineszenz Imaging Techniken für die nicht-invasive Evaluation von Strategien, welche die Infiltration von Melanomgewebe mit adoptiv transferierten T-Zellen und die anschließende Rekrutierung von Tumor-fördernden myeloiden Immunzellen modulieren; und (iii) Erweiterung des Modellsystems und Exploration von Behandlungsprotokollen die adoptive CD8+ T-Zell Therapien mit einer Hemmung der BRAF(V600E) Signaltransduktion kombinieren. Die geplanten Experimente sollen grundlegende Einsichten in Möglichkeiten der Kombination von T Zell Immuntherapien mit Hemmstoffen von Tumorwachstums-fördernden Signaltransduktionswegen ergeben und wertvolle Hinweise für aktuelle und zukünftige klinisch-translationale Studien für Patienten mit metastasierendem Melanom liefern.

Exploiting MET oncogene addiction in the HgfxCdk4R24C murine melanoma model to study therapy resistance to multimodal immunotherapeutic strategies
Laufzeit: 01.07.2014 bis 30.06.2018

Metastatic malignant melanoma is a highly aggressive and chemoresistant skin cancer with poor clinical outcome. Significant breakthroughs in the treatment of this devastating disease have been achieved in the recent years. Small molecule inhibitors against the mutated BRAF or MEK kinases and antibody-mediate blockade of negative immune checkpoint molecules like CTLA-4 and PD-1/PD-L1 prolong overall survival rates, but acquired resistance to these targeted therapies is the major obstacle for long-term remissions despite profound initial responses. Recent studies including ours identified diverse mechanisms of resistance to signaling inhibitors and immunotherapies in melanoma. We found that phenotypic plasticity of melanoma cells caused by a proinflammatory tumor microenvironment represents a critical route to resist a targeted T-cell immunotherapy directed against melanocytic antigens through reversible dedifferentiation without the need for acquired secondary hardwired genetic aberrations (Landsberg, , Hölzel, Tüting. Nature 2012). In this project we aim to scrutinize our central hypothesis that rational multimodal regimens combining targeted signaling inhibition with immunotherapeutic approaches represent a strategy to combat genetic and non-genetic tumor heterogeneity to achieve more durable responses in melanoma.To accomplish this goal we exploited oncogene addiction to the MET receptor tyrosine kinase in our HgfxCdk4R24C murine melanoma model and implemented CRIPSR/Cas9 genome engineering to establish a rapid modular pipeline to probe clinically relevant determinants of responsiveness to multimodal immunotherapeutic regimens. We hypothesize that targeted inhibition of oncogenic MET signaling synergizes with a T-cell therapy directed against the gp100 melanocytic antigen and we will explore how phenotypic plasticity and genetically engineered gp100 loss variants interplay to resist this immunotherapy. We expect to provide novel insight into the dynamic evolution of genetic and non-genetic melanoma heterogeneity in a preclinical therapeutic in vivo model of combined MET signaling inhibition and immunotherapy. Our long-term goal is to delineate new strategies that can be translated to improved clinical care. As MET plays a critical role in other cancers like hepatocellular carcinoma and lung cancer, we are convinced that our findings will have implications beyond melanoma.

Lokale Kontrolle von CD4+ T Zell Effektorfunktionen in Melanomen
Laufzeit: 01.01.2014 bis 31.12.2017

The skin immune system plays an important role in the development of malignant tumors. It can both inhibit and promote skin tumor growth. CD4+ T cells play an important role in the regulation of melanoma immunity and can exert potent antitumor immunity on their own. To study the role of CD4+ T cells we established a  TCR-transgenic mouse strain specific for the melanocytic antigen TRP1. Adoptively transferred TRP1-specific CD4+ T cells can cause autoimmune destruction of melanocytes and regression of skin melanomas in the context of prior lymphocyte depletion and adjuvant innate immune stimulation. However melanomas relapse early. The principal goal of our project in the next funding period is to understand how melanocyte-specific CD4+ T cell functions are regulated in primary cutaneous melanomas using this experimental system. The planned work is based on the central hypothesis that the ability of CD4+ T cells to cause autoimmune destruction of melanocytes and regression of primary cutaneous melanomas can be enforced by promoting effector functions of the Th17-Th9-Th1 spectrum of phenotypes. We anticipate that TRP1-specific CD4+ T cells normally shift towards exhausted, memory and regulatory phenotypes in the tumor microenvironment which then support tumor progression and angiogenesis. In this project we will first examine the phenotypic plasticity of CD4+ T cells by restimulating them with immunogenic or tolerogenic dendritic cells in vitro and in vivo (Aim 1). We will then establish strategies involving immunostimulatory nucleic acids and RNA aptamers to locally activate innate anti-viral immune pathways and re-direct dendritic cell functions which maintain anti-tumor effector functions of CD4+ T cells (Aim 2). We will use genetic tracing techniques to follow the fate of adoptively transferred CD4+ T cells in vivo, visualize their migration towards and adhesion to dendritic cells and melanoma cells, and evaluate their heterogeneity under different conditions  (Aim 3). These experiments will provide important insights how effector functions of CD4+ T cells could be harnessed for the treatment of melanoma.

Aktuelle wissenschaftliche Schwerpunkte
Rolle des Immunsystems und der zellulären Plastizität in der Pathogenese und Therapie des Melanoms:
Einfluss proinflammatorischer Mediatoren und Signalwege auf die Heterogenität und dynamische Plastizität von Tumor- und Immunzellen im Mikromilieu primärer und metastasierender Melanome. Bedeutung für die die lokale Regulation der Effektorfunktionen von melanom-spezifischen CD8+ und CD4+ T-Zellen. Auswirkungen auf die Tumorimmunüberwachung, die Tumorprogression und die Therapieresistenz. Kombination von tumorimmunologischen und tumorbiologischen experimentellen Ansätzen in genetischen Melanommodellen der Maus.
 
Entwicklung innovativer kombinatorischer Ansätze in der Tumortherapie:
Präklinische und klinische Entwicklung von effektiven Protokollen für die Kombination komplementärer Ansätze der Tumorimmuntherapie mit einem Fokus auf die Modulation von Entzündungs-getriebenen protektiven und regenerativen Vorgängen als Ursache für die Therapieresistenz. Lokale und systemische Stimulation von Rezeptoren für virale Nukleinsäuren (TLR/Helikasen) und Typ I IFN  mit rekombinanten viralen Vektoren und immunstimulierenden Oligonukleotiden, Kombinationstherapien mit immunmodulatorischen Antikörpern und Signaltransduktions-Inhibitoren.
